Microorganisms such as Cryptosporidium are enumeratedby techniques that are susceptible to considerablelosses, resulting in highly variable recoveries. Thisresearch quantified the methodological uncertainties inmicrobial concentration and removal data to demonstratethe effects of organism count, sample volume,replication, and methodological recovery on methodologicaluncertainty in microbial data. The authorsfound that the uncertainty associated with microbiologicalmethods is more substantially reduced by increasingorganism count to 10 or more than by improvingmethodological recovery.This investigation demonstrates the necessity oftreatment technique-based regulations such as the LongTerm 2 Enhanced Surface Water Treatment Rule. Findingsfurther suggest that before revisions to the TotalColiform Rule are finalized, a rigorous quantitativeanalysis should be conducted of the uncertainty associatedwith compliance data derived from the rule'sapproved analytical methods.
